SLU-PP-332 is a small molecule agonist of estrogen-related receptor alpha (ERRα), a master transcriptional regulator of mitochondrial biogenesis and oxidative metabolism. Unlike hormonal estrogen receptor ligands, ERRα is an orphan nuclear receptor that controls expression of genes governing fatty acid oxidation, oxidative phosphorylation, and muscle fiber type specification. SLU-PP-332 has been characterized as an exercise mimetic, reproducing key molecular signatures of physical training — including enhanced endurance capacity and metabolic reprogramming — without actual physical activity in preclinical models.
